I am doing a water pump change out and replacing all timing parts as recommended the question I have is I made sure the motor was at cylinder 1 tdc with all timing marks lined up I am using cam gear locks to hold them an place I reading the manual it says you turn the crank gear back one tooth when putting back on the timing belt do I do this with the locks in

Yes, the slack in the belt will come from the back of the engine and everything should line up when tensioned. Just turn the motor over a couple of rotations to check alignment.
